Fish have become an important model for studying hair cell plasticity and development. Unlike mammals, the inner ears of fish continue to grow and add hair cells throughout their lifespan, and fish can regenerate hair cells after inner ear damage. The plainfin midshipman fish (Porichthys notatus) is a useful model for investigating auditory plasticity because, in addition to these more widely studied contexts of hair cell plasticity, the midshipman auditory system undergoes changes associated with the reproductive state. In chapter 1, I summarize the multiple contexts of hair cell plasticity in fish and the secondary adaptive functions of the gas-filled swim bladder as an auditory and vocal structure. I then discuss the specific questions explored in this dissertation regarding developmental and reproductive state-dependent changes to the auditory inner ear and swim bladder using the plainfin midshipman as a model. In chapter 2, I provide evidence for a reproductive state-dependent change in hair cell density and a possible cellular mechanism in vocal type I male midshipman. In chapter 3, I describe changes in sensory epithelium area, hair cell density, and total hair cell quantity in the auditory inner ear throughout ontogeny in plainfin midshipman fish. In chapter 4, I discuss a change in the morphology of the swim bladder and its proximity to the inner ear in type I males related to reproductive state, indicating a possible change in adaptive use as an auditory structure to a vocal organ. Lastly, in chapter 5, I discuss the implications of the findings in this dissertation and future directions for studying auditory-related questions in the plainfin midshipman fish.

The reception and processing of socially relevant auditory signals is crucial for the reproduction and survival of many species including the plainfin midshipman fish, Porichthys notatus. Numerous behavioral, physiological and neuroanatomical studies have made P. notatus a robust model system for studying vocal acoustic communication. The ways in which fish are able to perceive acoustic information varies across species and I will detail the underlying morphological structures, including the swim bladder, that fish have adapted to enhance underwater hearing. I characterize the morphology of the midshipman swim bladder chapter two, in each of the three sexual phenotypes; females, type I and type II males. Using micro-computerized tomography I measure and quantify the swim bladder’s shape and relative distance to the inner ear organs in each midshipman sexual phenotype. In chapter three, using the immediate early gene cFos as a marker for neuronal activation, I quantify the response properties of gravid female midshipman to both conspecific advertisement calls and heterospecific white seabass calls. Specifically, I demonstrate the brain areas differentially activated during the perception of these socially relevant acoustic signals. To assess the role of the swim bladder in the neural processing of acoustic signals, in chapter four I conducted playback of conspecific advertisement calls to reproductive females with their swim bladders experimentally removed. Finally, in chapter five, I summarize the conclusions of my work as well as the broader implications and suggestions for future studies.

The plainfin midshipman, Porichthys notatus, has become a model for vocal communication and auditory reception because acoustic communication plays an important role in spawning behavior in this fish species. The auditory system of the teleost fish, P. notatus, is an important sensory receiver system used to encode intraspecific social communication signals in adults, but the response properties and function of this receiver system in pre-adult stages are less known. I describe what is known about the ontogeny of auditory sensitivity in juvenile and larval stages as well as other important background information in chapter one. In chapter two, I examine the response properties of auditory evoked potentials from the midshipman saccule, the main organ of hearing in this species, to determine whether the frequency response and auditory threshold of saccular hair cells to behaviorally relevant single tone stimuli change during ontogeny. I address the question of auditory sensitivity during larval development in chapter three and discuss when in the larval stage audition begins using the innate acoustically evoked behavioral response (AEBR) as a hearing assay. Specifically, I demonstrate that auditory reception develops early in larval development and that auditory sensitivity is similar throughout all larval stages. Additionally, during ontogeny, the saccule undergoes profound growth. I examine the saccular epithelia of larval, juvenile, and adult plainfin midshipman in order to describe the morphological changes that occur in saccular ultrastructure during development. These results are described in greater detail in chapter four. The results from chapters two through four are summarized and placed into broader contexts and I suggest future studies in chapter five.

This Technical Report presents the outcome of a Working Group that was established to determine broadly applicable sound exposure guidelines for fishes and sea turtles. After consideration of the diversity of fish and sea turtles, guidelines were developed for broad groups of animals, defined by the way they detect sound. Different sound sources were considered in terms of their acoustic characteristics and appropriate metrics defined for measurement of the received levels. The resultant sound exposure guidelines are presented in a set of tables. In some cases numerical guidelines are provided, expressed in appropriate metrics. When there were insufficient data to support numerical values, the relative likelihood of effects occurring was evaluated, although the actual likelihood of effects depends on the received level. These sound exposure guidelines, which are based on the best scientific information at the time of writing, should be treated as interim. The expectation is that with more research the guidelines can be refined and more cells in the tables completed. Recommendations are put forward defining the research requirements of highest priority for extending these interim exposure guidelines.
