In the application of delivery of therapeutics, nanostructures of various composition have been employed due to their capacity to act as a host for lipophilic payloads. Advances in the synthetic preparation, size, morphology, and chemical or physical characteristics of polymers have impacted their development and versatility. A detailed understanding of polymeric nanoparticle host-guest properties is crucial to their practical translation to specific delivery applications. Further, these features must be highly tailorable to overcome biological barriers, stably encapsulate their therapeutic contexts, and exhibit payload release selectively in the target environment. In this dissertation, we aim to rationally design polymeric nano-scale assemblies with well-defined compositions and unique stimuli-responsiveness to achieve desirable host properties and interfacial characteristics. While each chapter is specific in focus, we expect that the fundamental findings in this dissertation to broadly impact principles in drug delivery, including nanoparticle biodistribution (chapter 2), hydrophobic delivery (chapter 3), stimuli-responsive "smart" materials (chapter 4), and polymeric composition development (chapter 5).

Stimuli responsive nanoparticles have gained significant interest in the drug delivery research. The essential goal of drug delivery is to improve the efficacy of drugs by increasing their stability and bioavailability. Small molecule hydrophobic drugs face limited bioavailability due to their poor water solubility. Biologic drugs on the other hand, lack bioavailability because of their degradation by various enzymes encountered during blood circulation. For a systemic delivery approach, depending on site of drug action, delivery vehicles must pass through several contrasting micro environments before delivering the cargo selectively at the target site. Therefore, it is essential that a delivery vehicle shields the encapsulated therapeutics until reaching the targeted site, thereby improving the bioavailability and efficacy. Considering these parameters, our group has developed a nanoparticle based delivery platform (nanogel) aiming for cytosolic drug release. In this thesis we address x some of the challenges associated with nanoparticle mediated tumor targeted drug delivery, by utilizing the nanogel platform. By incorporating the novel nanoparticle design principles, we demonstrated efficient targeted delivery of hydrophobic drugs, as well as developed a new platform for delivery of hydrophilics. Additionally, we have also generated useful insights in the field of stimuli response polymeric nanoparticles which will be critical for future research in the field.

Advanced cancer therapeutic treatment strategies are rapidly evolving due to a better fundamental understanding of cancer cell biology and the complex tumor microenvironment. The development of tailored nanomedicine as a delivery platform for synergistic combination cancer treatment has grown in popularity over single faceted traditional chemotherapeutic or immunotherapeutic strategies. Chemotherapy and immunotherapy combined can target the heterogeneous cancer tumor from multiple pathways to augment the antitumor efficacy and prevent tumor metastasis and reoccurrence. Nanomedicine can be utilized synergistically to amplify antitumor immune responses of immunotherapy and minimize off-target toxicities of chemotherapy. However, the combination of multiple therapeutic strategies also compounds the obstacles that need to be overcome, such as the sequential biological barriers that the nanoplatform faces during delivery. Thus, the goal of this project was to develop a delivery platform that will facilitate the intracellular delivery of multiple cancer therapeutic agents for the treatment of colorectal cancer. Biodegradable, cationic nanogels were synthesized via a UV-initiated free radical emulsion polymerization to impart environmentally responsive behavior for the intracellular delivery of loaded therapeutics. The 2-(diethylamino) ethyl methacrylate-based copolymeric nanogels exhibited pH responsive behavior that was optimized for the pH gradient of the tumor microenvironment and endosomal compartments of cancer cells. Then, the cationic nanogels were assessed for their ability to load and release various chemotherapeutic and immunotherapeutic agents. Finally, the copolymeric nanoparticle system was evaluated in vitro for biocompatibility, cellular internalization, and therapeutic efficacy. This combination treatment strategy will provide an excellent foundation for the rational design and development of the next generation of drug delivery systems
