Years ago when we were asked to write a book on the present-day knowledge of the molecular biology of poliovirus, we did not expect that such an apparently simple task could involve so much time and effort. Our writing was hampered by the fact that both of us are full time "workers", so that this monograph is mainly a spare time expedience. The main attention of this book focuses on a detailed review of the molecular biology of poliovirus and especially on the advances of the last decade; medical and environmental aspects are only briefly mentioned. Observations from older studies are considered in view of more recent information. Some of the older ob servations provided fundamental insights and paved the way for present day research; too often such data has been neglected or independently rediscovered. Today, poliovirus research has again attracted considerable interest. It is hardly necessary to define the concept of receptors to readers of this series, but it should be borne in mind that in several instances receptors are undefined entities, whose molecular details remain to be established. On the other hand the ligand, which recognizes the receptors, has been identified and characterized in most cases. The current interest in the structure and function of biological membranes gives great expectations that we may understand in the near future the details of ligand-receptor interaction. This interaction involves two defined steps: the first, usually referred to as recognition, is followed by the second step, transduction, in which the ligand-receptor interaction is translated by the cell into a biochemical action. The present two volumes which cover prokaryotic and eukaryotic virus receptors, have been published together in order to illustrate the specificity of virus receptor recognition which appears to be a guiding principle for both bacteria and higher cells. The identification and characterization of the receptors for phages of gram-negative bacteria has to a large extent relied on the genetic tech niques available for these organisms. In a similar way the availability of genetic systems has also clarified the interrelationship between animal retrovirus receptors even if the molecular structure remains to be determined. The paucity of defined genetic systems may therefore explain part of our ignorance concerning the mole cular details of virus receptors on human cells and possibly also on gram-positive bacteria.

Immunopharmacology: A New Discipline of Immense Potential Among the looming triumphs of the biologic revolution is the rapidly developing understanding of the mechanisms of bodily defense. In the short span of 35 years, knowledge of immunologic machinery has progressed from crudest description to major understanding in cellular and molecular terms. Antibodies, immunoglobulins, and the complement system have been almost completely defined in detailed molecular terms. Organs, like thymus, spleen and lymph nodes-so long enigmatic black boxes-are beginning to be understood not only in cellular terms but in molecular, physiologic, and endocrinologic terms. With this surging new information about the immune system comes the possi bility of developing a pharmacology which can modulate and control immunologic functions. Immunopharmacology most broadly conceived must address (1) control of development and function of the cellular components of the immunologic appara tus; (2) facilitation and suppression of function of the immunologically competent cells of the several subclasses, like T helpers, suppressors, and effectors, and B effectors and suppressors; (3) manipulation and repair of the major biologic amplifi cation systems, e. g. , the complement system and kinin-kallikrein system, and (4) utilization, modulation, and inhibition of the galaxy of molecules generated by T lymphocytes, the lymphokines. This new pharmacology must deal with the funda mental effector mechanisms of immunity, namely inflammation, phagocytosis, vascular reactivity, and blood coagulation. Furthermore, immunopharmacology must address and manipulate cell-cell communication and interaction, so vital to control of the immunological apparatus.
