Volume 5 of Biomembranes covers an important group of membrane proteins, the ATPases. The P-type ATPases couple the hydrolysis of ATP to the movement of ions across a membrane and are characterized by the formation of a phosphoyrlated intermediate. Included are the plasma membrane and muscle sarcoplasmic reticulum Ca2+ -ATPases, the (Na+ -K+) -ATPase, the gastric (H+ -K+) -ATPase, the plasma membrane H+ -ATPase of fungi and plants, the Mg2+ - transport ATPase, the Salmonella typhimurium, and the K+ -ATPase of Escherichia coli, KdpB. The other important classes of ATPase in eukaryotic systems are the vacuolar H+ -ATPases and the F0F1 ATP synthase, and, in bacteria, the anion-translocating ATPases, responsible for resistance to arsenicals and antimonials, and the (Na+ -Mg2+) -ATPase of Acholeplasma. Finally, eukaryotic systems contain a variety of ectonucleotidases important, for example, in hydrolysis of extracellular ATP released as a cotransmitter from cholinergic and adrenergic nerve terminals. Volume 5 of Biomembranes explores structure-function relationships for these mebrane-bound ATPases.

The plasma membrane forms the living barrier between the cell and its surroundings. For this reason it has a wide range of important functions related to the regulation of the composition of the cell interior and to com munication with the cell exterior. The plasma membrane has therefore attracted a lot of research interest. Until the early 1970's it was only pos sible to study the plasma membrane in situ, its structure e. g. by electron microscopy and its function e. g. by uptake of radioactively labeled com pounds into the intact cell or tissue. The first isolation of plant protoplasts by enzymatic digestion of the cell wall in the early 1970's was an important step forward in that it provided direct access to the outer surface of the plasma membrane. More importantly, T. K. Hodges and R. J. Leonard in 1972 published the description of a method by which a fraction enriched in plasma membranes could be isolated from plant tissues using sucrose gradient centrifugation. As a result, the 1970's saw a leap forward in our understanding of the structurc and function of the plasma membrane. In 1981, S. Widell and C. Larsson published the first of a series of papers in which plasma membrane vesicles of high yield and purity were isolated from a wide range of plant tissues using aqueous polymer two-phase parti tioning.

In 1976 I wrote a monograph on lysosomes (Lysosomes: A Survey, Springer Verlag, Vienna) that was intended as an up-to-date, comprehensive survey. Whatever success I may have achieved then in fulfilling that intention, even the effort now would be foolhardy. The literature has grown so rapidly in the past decade that I certainly could not even read all of the essential papers, let alone understand and analyze them. My goal here, therefore, is simply to introduce the major features of lysosomes at a level I hope will be useful both to I;ldvanced students and to researchers interested in obtaining a broad background. This is in keeping with the design of the Cellular Organelles series: the series is more a set of advanced texts than of review monographs. This design carries with it the decision not to support each point by refer ences to the original literature. I apologize for the injustice involved in such a decision but feel that in any event it would be impossibly unwieldy to cite, adequately and in a balanced manner, the contributions of the vast network of researchers responsible for the information upon which I draw.
