Bambara groundnut is a drought-tolerant African legume. In order to estimate biodiversity, a DArT genotyping array was developed from which 296 unique markers were obtained. It was shown that the cultivated form was characterised by a generally narrow genetic basis and further genetic bottlenecks occurred with the dispersal of germplasm. Through 454 pyrosequencing of eight cDNA populations from contrasting genotypes and different treatments and a cDNA microarray constructed from selected transcripts, a complex network of gene regulation in leaves under water-deficit was described. In addition, two potentially novel candidate genes for drought-tolerance were identified. The approaches presented in this thesis could serve as an example to produce significant population genetic and transcriptomic data for other under-researched crops in a time- and cost-efficient way.
