Des méthodologies innovantes semblent être nécessaires pour mieux détecter l'origine des polluants, et pour la compréhension des processus physiques impliqués dans leur évolution dans le milieu. En outre, il est indispensable de vérifier si le traitement conduit à une minéralisation complète (efficacité du nettoyage). La mesure isotopique est un outil fondamental pouvant être utilisé dans la gestion d'une pollution : confinement, propagation et possibilités de bioremédiation est proposée dans le projet ISOTO-POL. En illustration de ce principe, le MTBE, fréquemment détecté dans les zones aquifères, est donné comme exemple. Les isotopes stables ont été utilisés avec succès pour démontrer la biodégradation naturelle ou améliorée des hydrocarbures pétroliers et additifs dans le sol et les eaux souterraines. Jusqu'à présent, seul le fractionnement isotopique total en 13C a été exploité, en utilisant la teneur en isotope mesurée par spectrométrie de masse de rapport isotopique (irm-MS). Mais cela conduit à une perte d'information importante concernant le site de la molécule concerné par la transformation. En revanche, la spectrométrie RMN offre la capacité de mesurer, site par site les rapports 13C/12C, donnant accès à de nouvelles informations. La méthodologie RMN isotopique permettra de déterminer les effets isotopiques position-spécifiques impliqués dans les transformations physiques (volatilisation et migration), chimiques (hydrolyse et oxydation) ainsi que la dégradation par les microorganismes. La construction d’un modèle fournira un moyen unique permettant la détection et le traçage détaillé d'un polluant donné, mais également la détermination de son origine
