Even though diffusion chamber culture was commenced long before ortho dox tissue culture by Metchnikoff (1887) there have been only sporadic attempts to use this methodology to study cell proliferation (review by Carsten, Chap. 1). Not so long ago diffusion chamber culture was nicknamed "confusion chamber" culture. I believe this conference has removed the confusion and will truly point out the intrinsic value of the system. It is not a substitute for established in vitro culture nor for in vivo studies. It complements both. Dr. Arne B~yum introduced diffusion chamber culture at Brookhaven National Laboratory. After some modest success in showing that one could culture human bone marrow and with appropriate stimuli induce erythro poiesis in diffusion chambers, several of the participants at this conference visited Brookhaven to learn firsthand this simple technology and to apply it in their own laboratories. However, the technique did not spread widely and controversy arose in which the same question was repeatedly asked: Can the diffusion chamber technique provide information that is not ob tainable more rapidly and easily, and at less expense by the in vitro tech niques? As a result of our deep interest in and involvement with diffusion chamber culture Dr. A. L. Carsten and I organized this conference. A major objective of this conference was to seek answers to the above question.

Normal murine, caprine, and human bone marrow and peripheral blood cells can be grown in diffusion chambers. The irradiated host provides a stimulus to erythrocytic and granulocytic growth over what is seen in the DC's implanted in the nonirradiated host. The stimulus is most likely humoral and affects erythropoiesis, granulopoiesis, and megakaryocytopoiesis. It is suggested but not proved that the stimulus for erythropoiesis is an increased level of plasma erythropoietin. It is tempting to suggest that CSF and thrombopoietin are responsible for the stimulation of the other cell lines. Growth of normal human cells in irradiated mice shows a marked reduction of the transit time from the myelocyte to the segmented neutrophils. Growth does not follow steady conditions in terms of rates and transit times. Since all cells produced are retained in the DC's after the time they would normally be extruded into the blood, a nonsteady state system exists. Senescent death of non-proliferating cells occurs. Human leukemic and multiple myeloma cells grow in DC's as do blood cells from patients with myelofibrosis and myeloid metaplasia. Data are insufficient to characterize aberrations in growth. Pilot studies show that human eosinophils will grow, proliferate and mature. (auth).

Normal human blood leukocytes were cultured in Millipore diffusion chambers implanted into the peritoneal cavities of irradiated mice. The evaluation of survival and proliferation kinetics of cells in lymphyocytic series suggested that the lymphoid cells are formed from transition of small and/or large lymphocytes, and the lymphoblasts from the lymphoid cells. There was also evidence indicating that some of the cells in these two compartments are formed by proliferation. The evaluation of plasmacytic series suggested that the plasma cells are formed from plasmacytoid-lymphocytes by transition, and the latter from the transition of lymphocytes. In addition, relatively a small fraction of cells in these two compartments are formed by proliferation. mature plasma cells do not and immature plasma cells do proliferate. Estimation of magnitude of plasma cells formed in the cultures at day 18 indicated that at least one plasma cell is formed for every 6 normal human blood lymphocytes introduced into the culture.
