Book Description
We demonstrate the feasibility of generating thousands of transgenic Drosophila melanogaster lines in which the expression of an exogenous gene is reproducibly directed to distinct small subsets of cells in the adult brain. We expect the expression patterns produced by the collection of 5,000 lines that we are currently generating to encompass all neurons in the brain in a variety of intersecting patterns. Overlapping 3-kb DNA fragments from the flanking noncoding and intronic regions of genes thought to have patterned expression in the adult brain were inserted into a defined genomic location by site-specific recombination. These fragments were then assayed for their ability to function as transcriptional enhancers in conjunction with a synthetic core promoter designed to work with a wide variety of enhancer types. An analysis of 44 fragments from four genes found that>80% drive expression patterns in the brain; the observed patterns were, on average, comprised of 100 cells. Our results suggest that the D. melanogaster genome contains50,000 enhancers and that multiple enhancers drive distinct subsets of expression of a gene in each tissue and developmental stage. We expect that these lines will be valuable tools for neuroanatomy as well as for the elucidation of neuronal circuits and information flow in the fly brain.

The Making and Un-Making of Neuronal Circuits in Drosophila

The Making and Un-Making of Neuronal Circuits in Drosophila PDF Author: Bassem A. Hassan
Publisher: Humana Press
ISBN: 9781493962396
Category : Medical
Languages : en
Pages : 278

Book Description
The small fruit fly, Drosophila melanogaster, has for over a century now had a large impact on biological and biomedical research; however, our knowledge of the fly brain has lagged significantly behind our understanding of other aspects of its development, physiology, and function. In The Making and Un-Making of Neuronal Circuits in Drosophila, innovative expert neuroscientists in the field present the ideas and concepts behind the methods, tools, and tricks that are currently being utilized to decode the secrets of this valuable insect’s brain. Focused on the concept of a neuronal circuit, defined as a series of synaptically connected neurons subservient to a particular behavioral modality, this volume contains chapters dealing with anatomical analysis with a focus on cellular and sub-cellular morphologies. These detailed approaches fall under the headings of “Physiology” and “Behavior”, conveniently divided the book into two sections. Written in the easy-to-follow Neuromethods series format, this work provides the kind of detailed description and implementation advice that is crucial for getting optimal results. Inventive and accessible, The Making and Un-Making of Neuronal Circuits in Drosophila provides the information and tools necessary to carry out current experiments and, more importantly, further advance the progress of the Drosophila neurobiology field and neurobiology in general.

Time, Love , Memory

Time, Love , Memory PDF Author: Jonathan Weiner
Publisher: Vintage
ISBN: 0804153361
Category : Biography & Autobiography
Languages : en
Pages : 322

Book Description
The story of Nobel Prize–winning discoveries regarding the molecular mechanisms controlling the body’s circadian rhythm. How much of our fate is decided before we are born? Which of our characteristics is inscribed in our DNA? Weiner brings us into Benzer's Fly Rooms at the California Institute of Technology, where Benzer, and his asssociates are in the process of finding answers, often astonishing ones, to these questions. Part biography, part thrilling scientific detective story, Time, Love, Memory forcefully demonstrates how Benzer's studies are changing our world view--and even our lives. Jonathan Weiner, winner of the Pulitzer Prize for The Beak of the Finch, brings his brilliant reporting skills to the story of Seymour Benzer, the Brooklyn-born maverick scientist whose study of genetics and experiments with fruit fly genes has helped revolutionize or knowledge of the connections between DNA and behavior both animal and human.

Decoding Neural Circuit Structure and Function

Decoding Neural Circuit Structure and Function PDF Author: Arzu Çelik
Publisher: Springer
ISBN: 3319573632
Category : Medical
Languages : en
Pages : 517

Book Description
This book offers representative examples from fly and mouse models to illustrate the ongoing success of the synergistic, state-of-the-art strategy, focusing on the ways it enhances our understanding of sensory processing. The authors focus on sensory systems (vision, olfaction), which are particularly powerful models for probing the development, connectivity, and function of neural circuits, to answer this question: How do individual nerve cells functionally cooperate to guide behavioral responses? Two genetically tractable species, mice and flies, together significantly further our understanding of these processes. Current efforts focus on integrating knowledge gained from three interrelated fields of research: (1) understanding how the fates of different cell types are specified during development, (2) revealing the synaptic connections between identified cell types (“connectomics”) using high-resolution three-dimensional circuit anatomy, and (3) causal testing of how iden tified circuit elements contribute to visual perception and behavior.

Nonlinear Circuits and Systems for Neuro-inspired Robot Control

Nonlinear Circuits and Systems for Neuro-inspired Robot Control PDF Author: Luca Patanè
Publisher: Springer
ISBN: 3319733478
Category : Technology & Engineering
Languages : en
Pages : 108

Book Description
This book guides readers along a path that proceeds from neurobiology to nonlinear-dynamical circuits, to nonlinear neuro-controllers and to bio-inspired robots. It provides a concise exploration of the essence of neural processing in simple animal brains and its adaptation and extrapolation to modeling, implementation, and realization of the analogous emergent features in artificial but bio-inspired robots: an emerging research field. The book starts with a short presentation of the main areas of the Drosophila brain. These are modeled as nonlinear dynamical structures, which are then used to showcase key features like locomotion, motor learning, memory formation, and exploitation. It also discusses additional complex behaviors, such as sequence learning and perception, which have recently been discovered to exist in insects. Much of the material presented has been tested in biorobotics classes for the Master’s degree in Automation Engineering and Control of Complex Systems at the University of Catania. Reporting on the work fostered by several national and international research projects, the book offers researchers novel ideas on how neuro-inspired dynamics can be used in developing the autonomous machines of the future.
