Book Description
Genetics more than any other biological approach can explain why some people experience more pain than others and receive less benefit from existing analgesics. Sixteen scholarly articles from international contributors describe the application of genetic techniques to the problem of pain and consider the knowledge that has so far resulted. Three themed sections review the techniques that are allowing the study of pain mechanisms at the genetic level; describe the progress being made in lab animals and humans in identifying the genes responsible for individual differences; and explore the practical and ethical issues that face pain researchers. Book Description
JOE BORLAND and Captain Beachboy deliver Lazarus team scientists to Bezo Metro Headquarters and find things in upheaval, so the rest of 9-Squad is ordered in to provide security. The Variant Squad has been shaken and bloodied over the last twenty-four hours with a rising body count, and there hasn’t been time for wounds to heal or for the dead recruits to be replaced. But the Bezo Board of Directors has put Lazarus specialists in charge of efforts to halt the Variant Effect outbreak in Metro, and end the Ziploc that holds the city in quarantine. The veterans Borland and Hyde are suspicious of the change in command but cannot override their questionable orders or abandon their squad to a dubious mission. As they prepare to join the scientists underground and open research labs that have been sealed for decades, 9-Squad finds the enigmatic company man, Brass, is working the graveyard shift and overseeing a top secret project of his own. Brass is in survival mode and his “Plan B” has hit a few bumps, but that’s not the worst of it. There have been reports of unusual sounds and sightings that lend some truth to the old rumors that the HQ basement levels are haunted. The squad must investigate the eerie phenomena, but all the signs point to something far more tangible and terrifying than the dead. Borland, Hyde and the frazzled 9-Squad must decide who they can trust as lethal forces gather, and avenues of escape begin to close. The only way to save their skins and protect the public from annihilation may involve a terrifying descent into darkness. Explore the MADHOUSE with Gas Light, second stage in the final chapters of The Variant Effect Series.

Book Description
THE VARIANT SQUAD barely had time to get back on its feet before the screaming started. Someone was getting skinned alive, and they knew it. But many of their comrades have just fallen to their deaths, and others are still missing, so Captain Beachboy knows that answering the cry for help would be suicide. Skin Eater packs are still hunting on the levels above and below them, and it’s only a matter of time before their ugly craving returns and they attack again. And that’s not the end of it. A murderous Kill Squad sent by an unknown enemy must have followed them underground. It has the Lazarus team’s innocent blood on its hands, and Beachboy suspects the unknown assassins are committed to spilling 9-Squad’s too. And the old myths about the basement are proving true. Something with an appetite for pain is haunting the bunker complex and feeds its hunger through the Biter packs. It has also poisoned the place with the Varion molecule that causes the lethal new form of the Variant Effect, forcing the squad to remain sealed in their bag-suits. And they’re running out of air. Variant Squad veteran Joe Borland knows that there are forces within the squad that might not want to win the battle underground, and would prefer leaving his bagged-comrades buried with the incriminating evidence they have found. So he’ll have to pick a side when his life’s on the line. In the meantime he and Beachboy take the squad survivors on a desperate search for a rumored escape tunnel while dark forces gather on all sides, and a secret clock winds down to doomsday and the final Variant Squad protocol of Ziploc, Gas and BURN! Explore the MADHOUSE to the END with BURN, third stage in the final chapters of The Variant Effect Series.

Chronic Postsurgical Pain

Chronic Postsurgical Pain PDF Author: Gérard Mick
Publisher: Springer Science & Business Media
ISBN: 3319043226
Category : Medical
Languages : en
Pages : 175

Book Description
Primum non nocere... The fact that a surgical procedure can leave any kind of pain casts a shadow over this tenet, which is seen as the basis of medical practice and anchor of its principle ethic... It is all the more surprising in that medicine has only paid attention to this paradoxical chronic pain situation for the past few years. Clarifying the knowledge acquired in this field has become all the more urgent for any care-giver today confronted by a legitimate request from patients: Why and how can a surgical procedure, which is supposed to bring relief, leave behind an unacceptable sequela? This is the approach which the contributors to this new subject of major clinical interest invite you to follow as you work your way through this book.

Pain Management and the Opioid Epidemic

Pain Management and the Opioid Epidemic PDF Author: National Academies of Sciences, Engineering, and Medicine
Publisher: National Academies Press
ISBN: 0309459575
Category : Medical
Languages : en
Pages : 483

Book Description
Drug overdose, driven largely by overdose related to the use of opioids, is now the leading cause of unintentional injury death in the United States. The ongoing opioid crisis lies at the intersection of two public health challenges: reducing the burden of suffering from pain and containing the rising toll of the harms that can arise from the use of opioid medications. Chronic pain and opioid use disorder both represent complex human conditions affecting millions of Americans and causing untold disability and loss of function. In the context of the growing opioid problem, the U.S. Food and Drug Administration (FDA) launched an Opioids Action Plan in early 2016. As part of this plan, the FDA asked the National Academies of Sciences, Engineering, and Medicine to convene a committee to update the state of the science on pain research, care, and education and to identify actions the FDA and others can take to respond to the opioid epidemic, with a particular focus on informing FDA's development of a formal method for incorporating individual and societal considerations into its risk-benefit framework for opioid approval and monitoring.

Organic Cation Transporter 1 (OCT1): Not Vital for Life, but of Substantial Biomedical Relevance

Organic Cation Transporter 1 (OCT1): Not Vital for Life, but of Substantial Biomedical Relevance PDF Author: Jurgen Brockmoller
Publisher: Frontiers Media SA
ISBN: 2889740293
Category : Science
Languages : en
Pages : 145

Book Description
Around one third of all biologically relevant small molecules are organic cations. These include endogenous substances like catecholamines and other neurotransmitters, toxins and drugs designed to affect signaling processes. The organic cation transporter 1 (OCT1) is among the strongest expressed membrane transporters at the sinusoidal (blood-facing) side of liver cells and contributes substantially to the clearance of the blood from numerous organic cations. A most striking feature of OCT1 is its pronounced genetic diversity. Between 1 and 10% of all human populations have little to no OCT1 activity. With several of the OCT1 substrates up to 10% of Europeans are functionally OCT1 deficient. Apparently, the lack of OCT1 do not lead to apparent substantial pathological changes in these individuals. It thus appears that this transporter is not essential to human life, but does it means that OCT1 is irrelevant? In the last 25 years since the first cloning of this transporter, data on its pharmacological and physiological relevance is steadily accumulating. Numerous clinically relevant drugs (e.g. metformin, morphine, fenoterol, sumatriptan, tramadol and tropisetron) have been shown to be substrates of OCT1, and OCT1 deficiency has been shown to affect the pharmacokinetics, efficacy, or toxicity of these drugs. Also vitamin B1 has been shown to be a substrate of OCT1, and in genetically modified mice OCT1 substantially modulated hepatic lipid metabolism, total body fat and systemic glucose and lipid concentrations. Still, numerous important questions remain unsolved: For which drugs, toxins, or other endogenous or exogenous substances is OCT1 relevant? How can we predict the relevance of OCT1 from in vitro studies? What determines the substrate selectivity of OCT1 in comparison to other transporters or transport processes for organic cations? What regulates the expression of OCT1 in the liver and possibly in other tissues? What is the impact of OCT1 variation in different areas of medicine, including the therapies for cancer as well as for pulmonary, cardiovascular, or neurological diseases? How can evolutionary biology contribute to a better understanding of the roles of OCT1? And, importantly, what types of research are likely to significantly further the knowledge on OCT1 in the next decades?
