Traffic jam (tj) encodes a soma-specific Maf transcription factor required for morphogenesis of the gonads in Drosophila melanogaster. I examined the requirement of tj function throughout oogenesis by studying the expression of follicle cell type specific markers in tj mutant cell clones. I also examined the interaction between tj and JAK-Stat pathway genes using a variety of loss- and gain-of-function mutations. As well as regulating the expression of a number of adhesion molecules, TJ appears to be a general requirement in follicle cells for their proper differentiation throughout oogenesis. Depending on when cells lose tj, their subsequent differentiation is blocked or defective. This suggests that like other Mafs, TJ is a regulator of differentiation. Furthermore, my findings suggest that TJ interacts with the JAK-Stat pathway during oogenesis. I have also identified a possible interaction between TJ and the EGF receptor signaling pathway.

The gene traffic jam (tj) encodes a soma-specific, large Maf transcription factor, required for gonad morphogenesis in Drosophila. My gain-of-function analysis of tj in adult ovaries revealed both, defects in cell specification and cell behaviour. The ectopic expression of tj during oogenesis produced follicles with ectopic polar cells, missing inter-follicular stalks and stalk cells, absent or non-migrating border cells, non-migrating centripetal cells and defective stretch cells. These tj gain-of-function phenotypes mimic the phenotypes found in Jak-Stat signalling pathway mutants. My data suggest that TJ may normally be involved in cell specification, and may interact with the Jak-Stat signalling pathway. It was also found that two of the mouse homologues of tj, MafB and c-Maf, are expressed in somatic tissues of adult mouse gonads, suggesting that the role of tj in gonad development and gametogenesis may be evolutionarily conserved.

In the follicle cell (FC) epithelium that surrounds the Drosophila egg, a complex set of cell signals specifies two fates that pattern the eggshell: the anterior centripetal FCs and the posterior columnar FCs. It has been shown previously that the long-range morphogen DPP represses the expression of the bunched (bun) gene in the centripetal FCs. Bun, in turn represses anterior gene expression and antagonizes Notch signaling to restrict centripetal FC fates in posterior cells. At stage 9, expression of the centripetal FC-associated gene slbo overlaps bun in anterior FCs; by stage 10A they repress each other's expression to establish a sharp slbo/bun expression boundary. The precises position of the slbo/bun expression boundary is sensitive to Notch signaling, which is required for both slbo activation and bun repression. As centripetal migration proceeds from stages 10B-14. slbo represses its own expression and both slbo loss-of-function mutations and overexpression approaches reveal that slbo is required to coordinate centripetal migration with nurse cell dumping. The expression of the transcription factor cut increases in the centripetal FCs at stage 10B in a pattern complementary to slbo. The pattern of cut expression results through slbo and cut cross-repression. Though Notch signaling represses cut expression prior to stage 7, Notch activation of slbo at 10A does not apparently influence the timing of cut expression at 10B. Notably, FLP-out clones expressing high levels of CUT anywhere in the columnar FCs are associated with increased accumulation of adherens junction proteins and noticeable apical constrictions so that cell shapes in CUT-expressing clones phenotypically resemble stage 10B centripetal FCs. Consistent with the notion that CUT is a centripetal FC selector gene, both cut mutant clones and FLP-out clones expressing slbo recovered the columnar FCs result in aberrant localization of apical adherens junction proteins to the basolateral locations. I propose that bun links long range morphogen signals to cell-contact dependent Notch signaling to produce a sharp columnar/centripetal cell fate boundary and that cut and slbo levels tightly regulate adherens junction polarity required for centripetal migration.
