Heterozygous Deletion of the FoxO1 Forkhead Transcription Factor Confers Protection Against High Fat Diet-induced Insulin Resistance

Heterozygous Deletion of the FoxO1 Forkhead Transcription Factor Confers Protection Against High Fat Diet-induced Insulin Resistance PDF Author: Hayley Baines

Book Description
Objective: Forkhead box O (FoxO) transcription factors represent evolutionarily conserved targets of insulin signaling, regulating metabolism and cellular differentiation in response to changes in nutrient availability. Although the FoxO1 isoform is known to play a key role in adipogenesis, its physiologic role in differentiated adipose tissue remains unclear. Research design and methods: In this study, we analyze the phenotype of FoxO1 haploinsufficient mice to investigate the role of FoxO1 in high fat-diet-induced obesity and adipose tissue metabolism. Specific assays include glucose and insulin tolerance tests, western blots, quantitative PCR, histologic analysis, and measurement of serum cytokines. Results: We show that reduced FoxO1 expression protects mice against obesity-related insulin resistance with increased PPAR[gamma] gene expression in adipose tissue and enhanced expression of known PPAR[gamma] target genes. Moreover, treatment of mice with the PPAR[gamma] agonist rosiglitazone improves in vivo insulin sensitivity in FoxO1 haploinsufficient animals, with reductions in circulating proinflammatory cytokines. Conclusions: These findings indicate that FoxO1 negatively regulates insulin action, and that this effect may be explained, at least in part, by inhibition of PPAR[gamma] activity.

New Transcription Factors and Their Role in Diabetes and Therapy

New Transcription Factors and Their Role in Diabetes and Therapy PDF Author: Jacob E. Friedman

Book Description
This book contains contributions from some of the most eminent experts in the fields of genetics, biochemistry, and pathophysiology of diabetes. Through specific examples, with broad applications, this book provides a comprehensive look at how transcription factors may underline the pathogenetic mechanisms of diabetes and obesity. Volume 5 provides an overview of the status of the field, while also providing valuable information of practical utility to those who do not necessarily work in this field. The integration of basic biology with physiologically and clinically relevant proteins should provide the reader with a theoretical background to understanding the strategies for new potential therapeutic targets and their application to disease. *Applies molecular biology to transcriptional regulation of metabolism and obesity*Underscores the clinical relevance of transcription factors*Provides a valuable overview of the current status of the field

mTOR in Human Diseases

mTOR in Human Diseases PDF Author: Olivier Dormond
Publisher: MDPI
ISBN: 3039210602
Category : Medical
Languages : en
Pages : 480

Book Description
The mechanistic target of rapamycin (mTOR) is a major signaling intermediary that coordinates favorable environmental conditions with cell growth. Indeed, as part of two functionally distinct protein complexes, named mTORC1 and mTORC2, mTOR regulates a variety of cellular processes, including protein, lipid, and nucleotide synthesis, as well as autophagy. Over the last two decades, major molecular advances have been made in mTOR signaling and have revealed the complexity of the events implicated in mTOR function and regulation. In parallel, the role of mTOR in diverse pathological conditions has also been identified, including in cancer, hamartoma, neurological, and metabolic diseases. Through a series of articles, this book focuses on the role played by mTOR in cellular processes, metabolism in particular, and highlights a panel of human diseases for which mTOR inhibition provides or might provide benefits. It also addresses future studies needed to further characterize the role of mTOR in selected disorders, which will help design novel therapeutic approaches. It is therefore intended for everyone who has an interest in mTOR biology and its application in human pathologies.
