Book Description
Serial analysis of gene expression (SAGE) was used to generate global gene expression profiles of 3T3-L1 preadipocytes and adipocytes. Our primary interest was in late gene expression, specifically genes that encode secreted proteins, so that we may detect novel adipocyte genes that are related to energy balance and obesity. SAGE detected the expression of apolipoprotein C-1 (apoC-I) mRNA in 3T3-L1 adipocytes, but not preadipocytes. ApoC-1, the product of the Apoc1 gene, is a 6.6 kDa protein which is found in plasma associated with chylomicrons, very low-density lipoproteins (VLDLS), and high-density lipoproteins (HDLs). The function of apoC-I and the regulation of its production is not well understood, though studies have suggested that it may interfere with triglyceride-rich lipoprotein clearance, free fatty acid transport into peripheral tissues, and cholesterol ester transfer protein (CETP) activity (Jong, et al., 1999). To elucidate the SAGE findings, the expression of apoC-I was determined by quantitative PCR and ELISA. Apoc1 mRNA expression was shown to increase during late-phase adipocyte differentiation and positively correlate with lipid accumulation, suggesting that it may be regulated by cellular triglyceride or cholesterol content. (Abstract shortened by UMI.).

The Adipocyte and Obesity

The Adipocyte and Obesity PDF Author: Charles H. Hollenberg
Publisher: New York : Raven Press
ISBN:
Category : Medical
Languages : en
Pages : 336

Book Description
Abstract: The proceedings of a 1982 international conference covers 33 review articles on research findings and the current state of knowledge of various metabolic and mechanistic aspects of the associations between the adipocyte fat cell and obesity. The articles address various facets of adipose cell growth and development; insulin action; lipid assimilation and lipoprotein and cholesterol metabolism in adipose tissue; lipolysis in adipocytes; and energy expenditure. The role of adipose tissue as a model system for the study of mechanisms of insulin action is highlighted by evidence of new cellular mediators. Recent insights in triglyceride assimilation regulation by lipoprotein lipase and fatty acid release through the lipolytic cascade are discussed. Other topics include the role of lipocytes in cholesterol turnover, the relationship of cholesterol metabolism to lipoprotein turnover and catabolism, and aspects of energy balance and thermoregulation in obesity. Mechanisms of Insulin Action

Mechanisms of Insulin Action PDF Author: Alan R. Saltiel
Publisher: Springer Science & Business Media
ISBN: 0387722041
Category : Medical
Languages : en
Pages : 223

Book Description
More than 18 million people in the United States have diabetes mellitus, and about 90% of these have the type 2 form of the disease. This book attempts to dissect the complexity of the molecular mechanisms of insulin action with a special emphasis on those features of the system that are subject to alteration in type 2 diabetes and other insulin resistant states. It explores insulin action at the most basic levels, through complex systems.

Molecular Nutrition and Diabetes

Molecular Nutrition and Diabetes PDF Author: Didac Mauricio
Publisher: Academic Press
ISBN: 0128017619
Category : Medical
Languages : en
Pages : 400

Book Description
Molecular Nutrition and Diabetes: A Volume in the Molecular Nutrition Series focuses on diabetes as a nutritional problem and its important metabolic consequences. Fuel metabolism and dietary supply all influence the outcome of diabetes, but understanding the pathogenesis of the diabetic process is a prelude to better nutritional control. Part One of the book provides general coverage of nutrition and diabetes in terms of dietary patterns, insulin resistance, and the glucose-insulin axis, while Part Two presents the molecular biology of diabetes and focuses on areas such as oxidative stress, mitochondrial function, insulin resistance, high-fat diets, nutriceuticals, and lipid accumulation. Final sections explore the genetic machinery behind diabetes and diabetic metabolism, including signaling pathways, gene expression, genome-wide association studies, and specific gene expression. While the main focus of each chapter is the basic and clinical research on diabetes as a nutritional problem, all chapters also end with a translational section on the implications for the nutritional control of diabetes. Offers updated information and a perspective on important future developments to different professionals involved in the basic and clinical research on all major nutritional aspects of diabetes mellitus Explores how nutritional factors are involved in the pathogenesis of both type1 and type2 diabetes and their complications Investigates the molecular and genetic bases of diabetes and diabetic metabolism through the lens of a rapidly evolving field of molecular nutrition
