A large amount of emphasis has been dedicated in recent years to introduce nanoparticles as a viable candidate for targeted therapies. In comparison to other candidates, mesoporous silica nanoparticles have the advantages of being biocompatible, easy to produce, and have the ability to prove to be a theranostic platform. To better study the specific targeting of diseased cells, adhesion studies of drug carrying bioconjugation constructs in fluid environments is required. The goal of this project was to develop a platform based on mesoporous silica nanoparticles that will be used for future multivalent adhesion studies. Specifically, mesoporous silica nanoparticles were synthesized with various sizes and aspect ratios, containing fluorescent dye to enable tracking studies, and with surface treatments that optimized stability and introduced primary-amine functional groups to facilitate attachment with targeted proteins and biomarkers. The effects of various parameters such as solvents, washing methods, secondary modifications and difference in concentrations of the starting materials for the synthesis mixture on the nanoparticles were studied in detail in this thesis. A portion of the study is also dedicated to optimizing a washing procedure to stabilize the particles in an aqueous medium in order to facilitate further modifications. The results of the work in this project can be utilized to provide a platform for further assays in flow chambers after bioconjugation with targeting proteins through orthogonal chemistries to study the adhesion properties in much greater detail.

This thesis involves synthesis, derivatization and biomedical applications of mesoporous silica nanopartilces (MSNs) and Fe3O4@SiO2 core/shell nanoparticles. Chapter 1 introduces the development of MSNs including the mesopores formation mechanism, synthesis conditions and their capability to act as stimuli responsive drug delivery platforms. In chapter 2, the synthesis optimization of different kinds of particles and their surface derivatization are introduced. Chapter 3 & 4 give specific examples of successful optimization and in vitro and in vivo application of MSNs enabled with pH-sensitive nanovalves and disulfide snap-tops for delivering the antibiotic moxifloxacin. It is shown that a high release capacity is necessary to reach a high efficacy ratio, compared with free drug. Chapter 5 discusses the uptake and release capacities of Fe3O4@SiO2 core/shell nanoparticles when modified with a pH-sensitive nanovalves, and its thermally cargo release behavior when surrounding temperature increases or an oscillating magnetic field is applied. In Chapter 6, successful distribution of Fe3O4@SiO2 core/shell nanoparticles in biofilms and on-command release of cargo inside biofilms are shown. Overall, these chapters demonstrate the ability of modifying both the outer surface and interior of MSNs, and their capability to act as a biocompatible controlled release platform that is more effective than equivalent amount of free drug.

The dissertation begins with Chapter 1, which is a general introduction of the fundamental synthesis of mesoporous silica materials, the selective functionlization of mesoporous silica materials, and the synthesis of nanostructured porous materials via nanocasting. In Chapter 2, the thermo-responsive polymer coated mesoporous silica nanoparticles (MSN) was synthesized via surface-initated polymerization and exhibited unique partition activities in a biphasic solution with the thermally induced change. In Chapter 3, the monodispersed spherical MSN with different mesoporous structure (MCM-48) was developed and employed as a template for the synthesis of mesoporous carbon nanoparticles (MCN) via nanocasting. MCN was demonstrated for the delivery of membrane impermeable chemical agents inside the cells. The cellular uptake efficiency and biocompabtibility of MCN with human cervical cancer cells were also investigated. In addition to the biocompabtibility of MCN, MCN was demonstrated to support Rh-Mn nanoparticles for catalytic reaction in Chapter 4. Owing to the unique mesoporosity, Rh-Mn nanoparticles can be well distributed inside the mesoporous structure and exhibited interesting catalytic performance on CO hydrogenation. In Chapter 5, the synthesis route of the aforementioned MCM-48 MSN was discussed and investigated in details and other metal oxide nanoparticles were also developed via nanocasting by using MCM-48 MSN as a template. At last, there is a general conclusion summarized in Chapter 6.
