Charles Ernest Overton's 1901 monograph Studien aber die Narkose has become a scientific classic in a number of different fields. This book represents the first English translation, and in fact the first translation into any other language, of the original German work. In addition to· the edited translation, this volume contains introductory chapters by Keith Miller, Peter Winter and Leonard Firestone and myself. As editor, I have attempted above all else to ensure that the translation faithfully represents Overton's ideas and data, while making the material readily understandable to the modem scientific reader. This has frequently required that extremely long sentences, common in turn-of the-century German but considered cumbersome today, be simplified into two or even three sentences. In addition, I have paid particular attention to the correct translation of scientific terms, and I accept complete responsibility for any inaccuracies in this area. Overton's original contents list included headings and subheadings, but only a fraction of these appear in the original text. For the sake of clarity they have all been included in the body of the translated work. Also included is an index containing all chemicals mentioned in the book, along with their Chemical Abstracts System Registry Numbers for un ambiguous identification, a complete list of Overton's publications (Appendix B), and a list of all biographical articles about Overton and articles dealing specifically with analyses of his data (Appendix C).

Neuropharmacology is a collection of papers presented at a symposium of the XXVI International Congress of Physiological Sciences, held in New Delhi, India in 1974. Contributors focus on the neurochemical action of central analgesics and their biochemistry, particularly Viminol R2 and azidomorphine, based on evidence derived using neurohistochemical techniques. This text is comprised of six chapters; the first of which deals with cholinergic mechanisms in narcotic analgesics. The riddle of the biochemical nature of drug dependence of the opiate type has stimulated many studies directed toward understanding the molecular basis of the action of opiates, and, particularly, the phenomena of tolerance, physical dependence, and drug-seeking behavior-phenomena exhibited by man and experimental animals exposed persistently to these drugs. The results of these studies provided a substantial body of information which has been published in the scientific and medical literature. The purely pharma cological responses in man and animals to the opiates have been described and evaluated in many monographs and text-books of pharmacology. However, there is no single source for specific and detailed information on the responses of the body and its tissues to narcotic analgesic drugs at the level of biochemical pharmacology; that is, the molecular history of the drug in the body and the biochemical consequences of its presence in tissue. This volume has been prepared in an effort to repair the deficiency. Two factors have contributed a special urgency to making this infor mation available in convenient form: (1) the current need for a better under standing of the biochemical mechanisms underlying addiction to narcotic drugs, and (2) the progress made in molecular biology which promises that significant advances in the elucidation of fundamental processes in the central nervous system and their drug-induced aberrations may soon be possible.
