Three experiments were conducted to determine the AID and SID of protein and AA in pea protein isolate (PPI), distiller's dried grains with solubles derived from a blend of wheat and corn (wcDDGS) and corn. In addition, 2 experiments were conducted to estimate the requirements of digestible SAA (dSAA) and digestible lysine (dLYS) for 3-week old broilers to obtain optimum growth performance(BWG, FCR), and optimum yield performance(CY, BMY and TDY). For digestibility studies, day-old male Ross 308 broiler chicks were fed test diets from day 1-14. The test-diets were balanced for vitamins and minerals and contained chromic oxide (0.3%) as an indigestible marker. The protein and AA digestibilities were determined either by direct or indirect method. For requirement studies, day-old male Ross 308 broiler chicks were fed lysine- or SAA deficient basal diets balanced for all other AA, except for the AA under test, according to the ideal protein concept. All test diets were also balanced for vitamins and minerals and were kept isonoitrogenous and isocaloric within each assay. The average SID estimates (%) of AA were 92.4, 71.5 and 93.3 in PPI, wcDDGS and corn, respectively. Using regression analysis, the dietary dLYS requirement estimates (%) were 1.12 and 1.13 for BWG and FCR, respectively. The dietary dSAA requirement estimate was 0.81% for BWG while no significant response was observed for FCR. Both CY and BMY increased cubically with increasing levels of dLYS, whereas, CY didnot respond significantly and BMY increased linerally with the increasing levels of dSAA.

The protein requirement for poultry is actually a requirement for amino acids. Recently, there has been much interest in formulating diets on a digestible amino acid basis. The most accepted methods of determining amino acid digestibility are the precision-fed cecectomized rooster assay (PFR) and standardized ileal chick assay (SID). The PFR involves tube-feeding cecectomized adult roosters and excreta are then quantitatively collected for amino acids. For the SID, 3-week-old broiler chicks are ad libitum fed a semi-purified diet with the test ingredient added as a sole source of protein for a period of several days. The animals are then euthanized and ileal digesta are collected and analyzed for amino acids. When these two methods were compared for 15 feed ingredients, standardized amino acid digestibility was found to vary among feed ingredients and among samples of the same ingredient. Amino acids (AAs) are not only building blocks of protein, but are also signalling molecules as well as regulators of gene expression and the protein phosphorylation cascade. Additionally, AAs are key precursors for syntheses of hormones and low-molecular weight nitrogenous substances with each having enormous biological importance. For example, physiological concentrations of AA metabolites (e.g., nitric oxide, polyamines, glutathione, taurine, thyroid hormones, and serotonin) are required for cell functions. Growing evidence shows that humans and animals have dietary requirements for all proteinogenic AAs. Mammals, birds and fish also have species- and age-dependent needs for some AA-related substances. However, elevated levels of other products (e.g., ammonia, homocysteine, H2S, and asymmetric dimethylarginine) are pathogenic factors for neurological disorders, oxidative stress, and cardiovascular disease. Thus, optimal amounts of AAs and their ratios in diets and circulation are crucial for whole body homeostasis and health. Adequate provision of one or a mixture of functional AAs or metabolites may be beneficial for ameliorating health problems at various stages of the life cycle (e.g., fetal growth restriction, neonatal morbidity and mortality, weaning-associated intestinal dysfunction and wasting syndrome, obesity, diabetes, cardiovascular disease, the metabolic syndrome, and infertility). Dietary supplementation of these nutrients can also optimize the efficiency of metabolic transformations to enhance muscle growth, milk production, and athletic performance, while preventing excess fat deposition and reducing adiposity. Therefore, functional AAs hold great promise in improving the growth, health and well-being of individuals.
