Book Description
Abstract: Optogenetics, utilising light-reactive proteins to manipulate tissue activity, are a relatively novel approach in the field of cardiac electrophysiology. We here provide an overview of light-activated transmembrane channels (optogenetic actuators) currently applied in strategies to modulate cardiac activity, as well as newly developed variants yet to be implemented in the heart. In addition, we touch upon genetically encoded indicators (optogenetic sensors) and fluorescent dyes to monitor tissue activity, including cardiac transmembrane potential and ion homeostasis. The combination of the two allows for all-optical approaches to monitor and manipulate the heart without any physical contact. However, spectral congestion poses a major obstacle, arising due to the overlap of excitation/activation and emission spectra of various optogenetic proteins and/or fluorescent dyes, resulting in optical crosstalk. Therefore, optogenetic proteins and fluorescent dyes should be carefully selected to avoid optical crosstalk and consequent disruptions in readouts and/or cellular activity. We here present a novel approach to simultaneously monitor transmembrane potential and cytosolic calcium, while also performing optogenetic manipulation. For this, we used the novel voltage-sensitive dye ElectroFluor 730p and the cytosolic calcium indicator X-Rhod-1 in mouse hearts expressing channelrhodopsin-2 (ChR2). By exploiting the isosbestic point of ElectroFluor 730p and avoiding the ChR2 activation spectrum, we here introduce a novel optical imaging and manipulation approach with minimal crosstalk. Future developments in both optogenetic proteins and fluorescent dyes will allow for additional and more optimised strategies, promising a bright future for all-optical approaches in the field of cardiac electrophysiology

Dynamic-Clamp

Dynamic-Clamp PDF Author: Alain Destexhe
Publisher: Springer Science & Business Media
ISBN: 0387892796
Category : Medical
Languages : en
Pages : 428

Book Description
Dynamic-clamp is a fascinating electrophysiology technique that consists of merging living neurons with computational models. The dynamic-clamp (also called “conductance injection”) allows experimentalists and theoreticians to challenge neurons (or any other type of cell) with complex conductance stimuli generated by a computer. The technique can be implemented from neural simulation environments and a variety of custom-made or commercial systems. The real-time interaction between the computer and cell also enables the design of recording paradigms with unprecedented accuracy via a computational model of the electrode. Dynamic-Clamp: From Principles to Applications contains contributions from leading researchers in the field, who investigate these paradigms at the cellular or network level, in vivo and in vitro, and in different brain regions and cardiac cells. Topics discussed include the addition of artificially-generated synaptic activity to neurons; adding, amplifying or neutralizing voltage-dependent conductances; creating hybrid networks with real and artificial cells; attaching simulated dendritic tree structures to the living cell; and connecting different neurons. This book will be of interest to experimental biophysicists, neurophysiologists, and cardiac physiologists, as well as theoreticians, engineers, and computational neuroscientists. Graduate and undergraduate students will also find up-to-date coverage of physiological problems and how they are investigated.

Basic Electrophysiological Methods

Basic Electrophysiological Methods PDF Author: Ellen Covey
Publisher: Oxford University Press
ISBN: 0199342547
Category : Medical
Languages : en
Pages : 241

Book Description
Basic Electrophysiological Methods provides a concise and easy-to-read guide on a selection of the most important contemporary electrophysiological techniques, their implementation, applications, and ways in which they can be combined and integrated with neuroscientific techniques. Intended for students, postdocs, and faculty with a basic neuroscience background, this text will not obscure the relevant technical details with textbook neuroscience tutorials as many other books do. Instead, each chapter provides a conscientious overview of the underlying theory -- a comprehensive description of equipment, materials, methods, data management, and analysis -- a troubleshooting guide, and a list of frequently asked questions. No book or online resource can function as strictly a DIY set of instructions on how to implement a complex technique. However, this book provides a fundamental and accessible set of information intended to form a foundation prior to, during, and after hands-on experience and training, greatly facilitating the initial learning process and subsequent fine-tuning of technical details.

Visualizing Chemistry

Visualizing Chemistry PDF Author: National Research Council
Publisher: National Academies Press
ISBN: 030916463X
Category : Science
Languages : en
Pages : 222

Book Description
Scientists and engineers have long relied on the power of imaging techniques to help see objects invisible to the naked eye, and thus, to advance scientific knowledge. These experts are constantly pushing the limits of technology in pursuit of chemical imagingâ€"the ability to visualize molecular structures and chemical composition in time and space as actual events unfoldâ€"from the smallest dimension of a biological system to the widest expanse of a distant galaxy. Chemical imaging has a variety of applications for almost every facet of our daily lives, ranging from medical diagnosis and treatment to the study and design of material properties in new products. In addition to highlighting advances in chemical imaging that could have the greatest impact on critical problems in science and technology, Visualizing Chemistry reviews the current state of chemical imaging technology, identifies promising future developments and their applications, and suggests a research and educational agenda to enable breakthrough improvements.
