L'allergie au latex est fréquente dans certains groupes à risque: le personnel de santé, les patients multi-opérés et les enfants atteints de spina bifida. Le latex présente treize allergènes reconnus par l'IUIS (International Union of Immunological Societies). Le caractère d'allergène majeur d'Hev b 2 et d'Hev b 13 et leur implication dans le syndrome latex-fruits font de ces 2 allergènes des candidats intéressants pour l'élaboration de formes recombinantes. Mon sujet de thèse est la caractérisation structurale d'Hev b 2 et Hev b 13 et de leurs épitopes B. Dans une première phase, les épitopes sont identifiés grâce à la technique SPOT couplée à un test de Coons indirect. Les structures tridimensionnelles sont ensuite définies par modélisation moléculaire par homologie. Les résidus composant les épitopes et localisés sur la surface du modèle tridimensionnel sont identifiés. Dans une deuxième phase, les premières étapes d'expression des allergènes recombinants sont réalisées.
