"The ability of enzymes to fold into unique structures drives their remarkable catalytic activity. Recently, a great effort is done in order to duplicate this high activity of natural enzymes by using simple molecules, for instance amphiphilic peptides, which can self-assemble into different structures in water, creating microenvironments that emulate the active site of enzymes. In addition, the controlled assembly of amphiphilic peptides governs their self-replicating propensity, as it promotes their own formation by self-recognition of building blocks, as was previously shown by our group. The addition of functional groups to the selfreplicating peptides may provide bi-functional systems, which will be able to selfreplicate and perform enhanced catalysis, benefiting one another (Figure 1). In this study, we have first used short dipeptides and further synthesized longer amphiphilic peptides that can readily self-assemble into elongated fibrils and catalyse hydrolysis of thioesters.. The supramolecular architectures formed by the selfassembly of the peptides was characterized using battery of techniques. The peptides were tested for the esterase-like activity using different substrates, and the difference between ester and thioester hydrolysis was tested. We observed that Fmoc-FH appears to catalyze the hydrolysis reaction of the thioester substrate better than the oxyester substrate. For control, we used molecules such as acetyl Histidine (aH) as a nonaggregating analogue. We found the the designed catalyst enhancing the catalysis as twice as fast than the control reaction. We were able to provide insights into the factors that dictate the efficiency of the catalytic reaction, and in particular to demonstrate the role of the self-assembly property upon the catalytic activity. We believe that this research will get us closer to the ability of assembling an autocatalytic-replication cycle that will carry out the both functions as well as benefiting each other. Finally, Chapter 5 illustrates the templating effect exerted by higher order complex, or self-assembling ligase. These results should contribute to not only to our understanding of nonlinear chemical systems, but also our ability to design de novo functional molecules especially peptides and proteins.
