Smad proteins mediate transforming growth factor-beta (TGF-beta) signaling to regulate cell growth and differentiation. SnoN is a negative regulator of TCF-beta signaling that maintains the repressed state of TGF-beta target genes in the absence of ligand. Upon TGF-beta stimulation, Smad3 and Smad2 translocate into the nucleus and induce a rapid degradation of SnoN, allowing activation of TGF-beta target genes. We have shown that Smad2- or Smad3-induced degradation of SnoN requires the ubiquitin-dependent proteasome and can be mediated by the anaphase promoting complex (APC) and the UbcH5 family of ubiquitin conjugating enzymes. Smad3 and to a lesser extent, Smad2, interact with both the APC and SnoN, resulting in the recruitment of the APC to SnoN and subsequent ubiquitination of SnoN in a destruction box- dependent manner. In addition to the destruction box, efficient ubiquitination and degradation of SnoN requires the Smad3 binding site in SnoN as well as key lysine residues necessary for ubiquitin attachment. Mutation of the lysine residues results in stabilization of SnoN and in enhanced antagonism of TGF-beta signaling. Our studies elucidate an important mechanism and pathway for the degradation of SnoN and more importantly, reveal a novel role of the APC in the regulation of TGFS signaling.

Scientists often look askance at their colleagues whose research appears too strongly focused on a single gene or gene product. We are supposed to be interested in the “big picture” and excessive zeal in pursuit of a single pixel might seem to border on an obsession that is likely to yield only details. However as this volume of Current Topics in Microbiology and Immunology demonstrates, this is certainly not the case for myc. Intense study of this en- matic proto-oncogene over the last twenty years has only broadened our view of its functions and led to insights into mechanisms relating to transcriptional regulation as well as to cell growth, proliferation, differentiation, apoptosis and organismal development. The myc gene originally came to light as a retroviral oncogene (v-myc) associated with a wide range of acute neoplasms. It was later shown to be a virally transduced cellular gene (c-myc) which is a member of family of on- genes (c-myc,N-myc,L-myc). These family members are themselves subject to a bewildering assortment of genetic rearrangements associated with many different types of tumors derived from many different types of cells. These rearrangements (including chromosomal translocation, viral integration, and gene ampli?cation) act to uncouple expression of the myc family genes from their normal physiological regulators. The chapter by LIU and LEVENS - scribes the key pathways leading to regulation of myc expression, showing that such regulation occurs at several different levels and through multiple mechanisms.

But when a character simply leaves out of selfishness or self-preservation, abandoning a child out in the yard on a swing set, the ramification can be life destroying.Some of these stories I've read in earlier incarnations and some are new to me, as they will be to the fortunate readers of this collection. "Squirrels" shows a widow becoming undone, heralding the end of her freedom of living alone. Her longing for her dead husband has driven her to attack, in funny and crazed ways, an unseen army of squirrels. She is convinced the squirrels want her out of her house. But her real evictor, her daughter, is more difficult to conquer. In "Visiting Day" we witness a vital elderly woman, who is strong and confident at the start, lose her way. All because the old sometimes have a way of reverting to acting like school children, and the simple action of sitting down at a table can have debilitating consequences. "Mirror, Mirror" is a nice reboot of a fairy tale, but there is no happily ever after for Mariella. The flash fiction, "Five", is one of my favorites in this collection. Regan is pitch-perfect in showing, through actions and dialogue, this emotional moment. A young child's birthday is set aside because of a down deep hurt. And more escapes ensue.In the longest piece in this collection, "Loose Threads", Anne navigates through the coming apart of her family after the death of her brother. Addictions sever the many ties between characters throughout this novella: from drugs, to abusive relationships, to victimization. In the end, though, there comes finally a kind of redemption by way of a baby's birth. Anne is reconnecting with a tether that may bring her back to where she deserves to be.This is Emily Regan's first book. And I'm certain it will not be her last. She has done something here that calls for attention. She has opened familiar wounds that we need to consider, but will not be able to readily heal. And, as many of us may admit, sometimes running away from what hurts us seems the easiest way out. Although, I must acknowledge that Regan, once a student of mine, is now instructing me with her wonderful writing and vivid characters that easiest may work for a moment, but the many moments, days, weeks, and years that follow tend to have a way of making life more and more difficult. Good writing, in my opinion, should present this outcome, and Emily Regan is a very good writer.-- Dan Crawley (from the Preface)
