Abstract : Lectins are carbohydrate or glycan binding proteins produced by every living species. Binding of lectins to glycan epitopes of glycoproteins is essential for initiating a variety of biological functions. Galectin-3 (Gal-3) is an endogenous human lectin that plays important roles in cell proliferation, adhesion, differentiation, metastasis, angiogenesis, and apoptosis. This dissertation describes two new binding properties of Gal-3 that are physiologically relevant. The new interaction of Gal-3 described herein significantly extends its biological function. Biological interactions of Gal-3 are known to occur when it binds to glycoproteins. However, we found that Gal-3 also binds to another class of glycoconjugates, namely, proteoglycans and their constituent glycosaminoglycans. Calorimetric data show that Gal-3 binds with glycosaminoglycans (heparin and chondroitin sulfate) and chondroitin sulfate proteoglycans (CSGPs). Spectroscopic data reveal that Gal-3 cross-links with chondroitin sulfate A and C as well as a CSGP and forms insoluble complexes. Such cross-linking of Gal-3 by glycosaminoglycans is potentially important for many biological processes including cell-signaling. We also discovered that Gal-3, produced by malignant thyroid gland, interacts with a thyroid cancer biomarker, thyroglobulin (Tg) with nanomolar affinity. This interaction can interfere with the clinical detection of thyroid cancer and can also impact thyroid hormone production and immune response. To produce sufficient Gal-3 for these studies, a new purification protocol named 'Capture and Release' (CARE) was developed. This protocol is cost-effective, faster and was found to be efficient enough to purify other lectins besides Gal-3. The function of lectins including Gal-3, depends on the interaction with their binding partners (glycoconjugates). Glycoconjugate is a common name of any macromolecule that contains glycans linked to a protein or lipid core (scaffold). The outcome of lectin-glycoconjugate interactions is believed to depend solely on the structures of the lectin and the glycan epitopes. However, we discovered that the scaffold part plays a vital role in the binding process. This dissertation thus describes a novel molecular mechanism associated with glycoconjugate functions and new physiologically-relevant recognition properties of an important human lectin (Gal-3).

Changes in serum glycosylation have long been associated with hepatocellular carcinoma (HCC), and these modifications are often detected with sugar-binding proteins called lectins. HCC is associated with increased serum fucosylation, and this sugar is often detected with the Aleuria aurantia lectin (AAL). However, this lectin binds fucose in several forms, one of which is associated with cancer ([alpha]1-6 linked fucosylation) and some of which are not. Thus, its use in clinical and tissue analysis is limited. We have created a recombinant AAL, which contains a single amino acid mutation: N224Q (rAALN224Q), within the binding site 5, and this lectin has increased sensitivity to the cancer-specific fucosylated glycan. Our analysis of rAALN224Q has shown that the mutation within one specific fucose binding pocket had a major impact upon the mutated site as well as on sites that were not mutated and led to a lectin with altered binding specificities. This lectin is currently part of a diagnostic test under evaluation for the early detection of HCC. We have further explored the changes in N-linked glycosylation associated directly with HCC tissue through a method of N-linked glycan matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI IMS). MALDI IMS of N-linked glycan revealed inter- and intra-tumor heterogeneity shown by expression and spatial localization of glycans in the tumor. Tumors were associated with alterations in mannosylation, fucosylation, branching, and complex glycan formation. Importantly, all tumors were associated with some change in glycosylation, but there was significant glycan heterogeneity.

I organized this symposium, "Molecular Immunology of Complex Carbohydrates-2 (MICC-2)," at the Institute of Biological Chemistry, Academia Sinica, Taipei, Taiwan th between August 28-September 2, 1999, as a satellite meeting of the 15 International Glycoconjugate Conference (held August 22-27 in Tokyo, Japan). I also held a Taiwan Canada Glycobiology Workshop after this meeting at the Institutes of Biological Chemistry and Chemistry. To promote glycobiology in Taiwan, I offered a Complexs Carbohydrate & Medicine-2 workshop at the Glyco- Research Laboratory, College of Medicine, Chang Gung University, Kwei-san, Tao-yuan, Taiwan before the MICC-2 symposium. The lecture and poster materials of these three meetings are collected in these proceedings, which are divided into five Sections and two Appendixes. Section I, entitled "Protein-Carbohydrate Interactions of Plant and Animal Lectins," provides current concepts of lectin-carbohydrate interactions; classification of lectins, based on amino acid sequences, molecular structures, and lectin affinity for carbohydrates. However, the relationships between amino acid sequences and carbohydrate affinity of lectins have to be more thoroughly characterized. The reviews on animal lectins in this Section explore new areas of lectins. Section II, "Aspects of Structure and Antigenicity of Glycoconjugates," provides important information on structural concepts of glyco immunology. "Glycotope Expression (Glycosylation), Metabolism and Functions," which play important roles in life processes, are discussed in Section III. Four articles on advances in knowledge on structural roles of glycans and treatment of cancer are discussed in Section IV.
