The Hepatocyte Review

The Hepatocyte Review PDF Author: M. N. Berry
Publisher: Springer Science & Business Media
ISBN: 9401733457
Category : Science
Languages : en
Pages : 597

Book Description
It is thirty years since the technique of high-yield preparation of isolated hepatocytes, by collagenase perfusion of the liver, was published. The original method described by Berry and Friend has undergone many minor modifications by other workers, and the two-step procedure introduced by Seglen in 1976 has become the most frequent way to prepare hepatocyte suspensions. An important development introduced by Bissell in 1973 was the use of the cells as the first step in monolayer culture. The availability of the isolated hepatocyte preparation as cells in suspension or culture has undoubtedly facilitated research on the liver. This was emphasised in our book, published (with Dr. Greg Barritt) in 1990, which described in detail methods of preparation and the properties of the isolated hepatocytes. It also discussed the usefulness of the preparation for the study of intermediary and xenobiotic metabolism, calcium ion transport, and the growth and differentiation of hepatocytes in culture. The book also touched briefly on a range of specialised techniques, including peri fusion, subcellular fractionation, transplantation, cryopreservation and measurement of intracellular pH. Although standard procedures for the manipulation of hepatocytes have not changed a great deal in ten years, they have undoubtedly been refined. This applies particularly to hepatocyte culture techniques, cryopreservation, and even to preparation of hepatocyte suspensions, where it is now feasible to use purified enzymes. There is also much more emphasis on the use and study of human hepatocytes, particularly in the field of pharmacology and therapeutics.

The Isolated hepatocyte

The Isolated hepatocyte PDF Author: E.J. Rauckman
Publisher: Elsevier
ISBN: 0323156797
Category : Medical
Languages : en
Pages : 305

Book Description
The Isolated Hepatocyte: Use in Toxicology and Xenobiotic Biotransformations covers the link between research on the isolated hepatocyte and the disciplines of cell culture, toxicology, metabolism, and molecular biology. This book is composed of 11 chapters and begins with an overview of the regulation of liver growth, sulfation, glucuronidation of xenobiotics in specific liver sites. The next chapters deal with toxicology studies in cultured hepatocytes from various species and the in vitro control of hepatocyte proliferation. These topics are followed by discussions of choices and methods of cytotoxicity measures; hepatoxicity of metals; the metabolism and toxicity of xenobiotics in a primary culture; and the mechanism of carcinogen-induced pleiotropic drug resistance in hepatocytes. The concluding chapters describe the in vivo and in vitro measurement of chemically-induced DNA repair in hepatocytes, as well as the genotoxicity studies with human hepatocytes. This book is intended primarily to toxicologists and researchers.

Applications of Pharmacokinetic Principles in Drug Development

Applications of Pharmacokinetic Principles in Drug Development PDF Author: Rajesh Krishna
Publisher: Springer Science & Business Media
ISBN: 1441992162
Category : Medical
Languages : en
Pages : 556

Book Description
This volume is an important advancement in the application ofpharmacokinetic (PK) and pharmacodynamic (PO) principles to . drug development. The series of topics presented deal with the application of these tools to everyday decisions that a pharmaceutical scientist encounters. The ability to integrate these topics using PK and PO methods has optimized drug development pathways in the clinic. New technologies in the areas of in vitro assays that are more predictive of human absorption and metabolism and advancement in bioanalytical assays are leading the way to minimize drug failures in later, more expensive clinical development programs. of Pharmacokinetics and pharmacodynamics have become an important component understanding the drug action on the body and is becoming increasingly important in drug labeling due to it's potential for predicting drug behavior in populations that may be difficult to study in adequate numbers during drug development. The ability to correlate drug exposure to effect and model it during the drug development value chain provides valuable insight into optimizing the next steps to derive maximum information from each study. These principles and modeling techniques have resulted in an expanded and integrated view of PK and PO and have led to the expectations that we may be able to optimally design clinical trials and eventually lead us to identifying the optimal therapy for the patient, while minimizing cost and speeding up drug development. There is wide utility for the book both as a text and as a reference.

Book Description
Advances in Tissue Banking is the only series of publications where producers and clinical users of tissues can learn in one place about developments in this interdisciplinary field. This important volume concentrates on the new leading topics, and bridges the tissue banking technology and the scientific developments, which fuel this rapidly growing field. It presents the regulatory controls being introduced in the USA, Europe and Japan, as well as the relationship among technical orthopaedics, in vitro tissue engineering, and osteoinduction. New subject areas in the volume include cell processing and preservation, particularly for islet and stem cells; preservation, processing and viability of vascular grafts; and gland transplantation. Collagen biochemistry is reviewed and linked to skin substitutes and skin banking. The volume fully reflects present global developments, with contributions from Spain, Japan, Russia, Poland, Estonia and the USA, among other countries.
