"Epidermal growth factor receptor (EGFR), a prominent oncogenic driver, has been a major target in the advent of personalized medicine and has elicited considerable success in the forms of antibodies and EGFR tyrosine kinase inhibitors (EKI). Extracellular vesicles (EVs) are membrane fragments that mediate short- and long-distance cellular communication by stably carrying biologically active cargo including proteins, lipids, and nucleic acids. EVs play key roles in cellular transformation, angiogenesis, and metastasis. This thesis explores the relationship between EKI treatment and extracellular vesicles as a conduit for potential unintended biological effects as a result of treatment with targeted agents. We observed cellular responses and examined vesiculation profiles in several cancer cell lines with varying EGFR mutation status and expression in response to second-generation EKI dacomitinib. We speculate that quantification of EVs reflects response to drug treatment, as cell lines sensitive to dacomitinib increase vesiculation while cells that are resistant do not. Further, we observed that EV-associated DNA content was relative to drug concentration in sensitive cells. Endothelial cells exhibited different responses when cultured in the presence of EVs derived from dacomitinib-treated cells and untreated cells, suggesting that EKI treatment modifies the angiogenic potential of released EVs. We postulate that, due to their physical characteristics and associated cellular responses at corresponding drug concentrations, EVs derived from EKI treatment belong to an emerging subtype of apoptotic exosome-like vesicles"--
