Measles, also called the greatest killer of children in history, still annually affects about 50 million individuals and causes close to a million deaths primarily in developing countries. Before the advent of measles vaccine some 30 years ago, these figures were roughly three times higher. Attenuated measles virus (MV) strains, all quite closely related to the original Edmonston isolate, have a very good record as a safe and highly efficacious vaccine and have brought down the measles toll in industrialized countries to almost negligible levels. However, recent outbreaks in the USA and Europe have again brought the measles problem to public attention. Sadly enough, these outbreaks were more instrumental in inducing activities to drastically reduce and hopefully finally eradicate measles than were the ten thousand times higher number of victims in developing countries. To reach this goal, as detailed in this volume, apparently it is not enough to of the existing vaccine as was the rigorously enforce use case with smallpox eradication: the intricacies of measles disease phenomena, in particular the generalized immune suppression which favors secondary infections, require more basic knowledge of the virus-host interactions and probably the development of new vaccines for special applications such as first immunizations of very young infants in developing countries.

Wolf's discovery demonstrating that a reporter gene is expressed in myocytes subsequent to injection of naked DNA, was exploited by immunologists and vaccinologists to develop a new generation of vaccines. This observation galvanized the research and in a short lapse of time, an oceanic volume of knowledge has been accumulated. The research carried out in a variety of animal models showed the efficacy of genetic immunization against viruses, bacteria, and some parasites by the ability to induce a strong priming effect resulting from long-lasting persistence of plasmid as episomes. Furthermore, it was demonstrated that newborn or infant immune unresponsiveness to classical vaccines can be corrected by genetic immunization. Immunologists have provided pertinent information on the cellular basis of the immune responses elicited by genetic immunization, and molecular biologists have established the molecular basis of intrinsic adjuvant properties of plasmids.

Innate immunity is one the most evolutionally conserved systems, designed to protect the organism from viruses and bacterial infections, stress and many other types of attacks from the outside world. During the past decade, the capacity of molecular biology and information technology to produce and analyse data have grown exponentially, rapidly reforming many aspects of immunology research in the post-genomics era. As a result, scientific understanding of signalling networks governing the innate immunity response in human tissues and other organisms has evolved beyond recognition, compared to even just a decade ago. Many strategies have been designed over the years to identify novel proteins, which have a crucial role in innate immunity responses by regulating particular signalling pathways. These projects had many advantages, including the definition of novel drug targets, as exemplified by the recent success of anti-TNF therapy, as well as leading to a better, system-wide understanding of the molecular control of innate immunity. In the past few years, a new concept, Immunomics, has been adopted to define an emerging, multidisciplinary field of research (Schonbach, 2003). Although rapid progress has been made to identify the proteins playing pivotal roles in the innate immunity–related signalling pathways (for example, TIR signalling pathways), the catalogue of proteins with a key regulatory function identified and studied is far from completed. Novel proteins need to be char- terised to gain a more comprehensive picture of how signalling networks are regulated.
