Vegetable legumes are largely cultivated by resource-poor rural households in Bangladesh for domestic consumption as a source of protein and micronutrients and to sell in local markets for family income. Sustainable cultivation of these crops in subsistence farming systems in Bangladesh is constrained by a wide range of biotic and abiotic factors, including viral diseases. This study was undertaken to (i) characterize Bean common mosaic virus (BCMV) from Rajma beans (RB, Phaseolus vulgaris), country beans (CB, Lablab purpureus) and yard-long bean (YLB, Vigna unguiculata (L.) Walp. ssp. sesquipedalis Verdc.) and Cowpea mild mottle virus (CPMMV) from CB and YLB and (ii) identify sources of resistance to the pea aphid (Acyrthosiphon pisum) in Lentil (Lens culinaris) germplasm. Total RNA from candidate samples were subjected to high-throughput Illumina sequencing to generate near full-length genome sequences of BCMV and CPMMV. Pairwise comparisons of genome sequences and phylogenetic analysis with corresponding viral sequences provided new insights on the genetic variability of BCMV and CPMMV in vegetable legume crops commonly grown by farmers. An analysis of pods from BCMV-and CPMMV-infected CB and YLB showed that virus-infected plants produce nutritionally poor-quality pods. Seed transmission studies have shown that seed lots from local markets can play epidemiologically important role in the field-spread of BCMV and CPMMV. The results will help to establish robust phytosanitary measures for the supply of certified seeds to smallholder farmers within Bangladesh. Experiments were conducted to identify genetic resources and genes associated with resistance to the pea aphid (Acyrthosiphon pisum) in 188 lentil (Lens culinaris) accessions of the USDA Single Plant Core Collection. A cluster analysis associated with the mean number of adults, nymphs and total aphids developed on lentil germplasm lines identified several lines with significant levels of resistance to the aphid. The genotyping by sequencing-based Single Nucleotide Polymorphism analyses were used for genome-wide association mapping of fifteen candidate genes or quantitative trait loci associated with aphid resistance. The genetic resources identified in this study is useful for breeding programs to improve resistance against the pea aphid as a pest and to reduce the spread of aphid-transmitted viruses.
