This book grew out of a Conference on "Genetic Consequences of Nu cleotide Pool Imbalance" which was held at the National Institute of Environmental Health Sciences on May 9-11, 1983. The Organizing Com mittee consisted of Drs. Diana Anderson (BIBRA), Robert Haynes (York University), Bernard Kunz (NIEHS), Barry Glickman (NIEHS), and Frederick J. de Serres (NIEHS), Chairman. The meeting was org- ized by the staff of the Office of the Associate Director for Genetics and was financed solely by the National Institute of Environmental Health Sciences. Heritable human genome editing - making changes to the genetic material of eggs, sperm, or any cells that lead to their development, including the cells of early embryos, and establishing a pregnancy - raises not only scientific and medical considerations but also a host of ethical, moral, and societal issues. Human embryos whose genomes have been edited should not be used to create a pregnancy until it is established that precise genomic changes can be made reliably and without introducing undesired changes - criteria that have not yet been met, says Heritable Human Genome Editing. From an international commission of the U.S. National Academy of Medicine, U.S. National Academy of Sciences, and the U.K.'s Royal Society, the report considers potential benefits, harms, and uncertainties associated with genome editing technologies and defines a translational pathway from rigorous preclinical research to initial clinical uses, should a country decide to permit such uses. The report specifies stringent preclinical and clinical requirements for establishing safety and efficacy, and for undertaking long-term monitoring of outcomes. Extensive national and international dialogue is needed before any country decides whether to permit clinical use of this technology, according to the report, which identifies essential elements of national and international scientific governance and oversight.

The finding that chemicals can be metabolically activated to yield reactive chemical species capable of covalently binding to cellular macromolecules and the concept that these reactions could initiate toxicological and carcinogenic events stimulated a meeting by a small group of toxicologists at the University of Turku, in Finland, in 1975 (Jollow et al. , 1977). The growing interest in this field of research led to subsequent symposia at the University of Surrey, in England in 1980 (Snyder et al. , 1982), and the University of Maryland in the U. S. A. in 1985 (Kocsis et al. , 1986). The Fourth International Symposium on Biological Reactive Intermediates was hosted by the Center for Toxicology at the University of Arizona and convened in Tucson, Arizona, January 14-17, 1990. Over 300 people attended. There were 60 platform presentations by invited speakers, and 96 volunteer communications in the form of posters were offered. These meetings have grown from a small group of scientists working in closely related areas to a major international series of symposia which convene every five years to review, and place in context, the latest advances in our understanding of the formation, fate and consequences of biological reactive intermediates. The present workshop had its orlglns in discussions among Alexander Hollaender, Oddvar Nygaard, Donald Borg, Richard Setlow and Victor Bond on the need for a symposium that would deal with a broad spectrum of pressing subjects related to the physics, chemistry, and biological actions of ionizing radiations, and the theoretical and practical problems of risk assessment. It soon became apparent that the spectrum of subjects considered was too broad for the desired depth of coverage; in fact, it seemed un likely that the conference participants would have the background knowledge to span the gamut. Therefore, two separate meetings were decided upon, the first of which, Comparison of Mechanisms of Carcinogenesis by Radiation and Chemical Agents, was held at the National Bureau of Standards, Gaithersburg on December 6-7, 1983. The meeting was sponsored by the NCI. The second meeting has emerged as the present workshop at Brookhaven National Laboratory. An interface between the two conferences has been provided by Michael Fry and Donald Borg, who have summarized the salient points emanating from the NCI Symposium. We intended that the first conference should focus on the basic mechanisms of radiation and chemical carcinogenesis, while the second, the present meeting, would emphasize exposure-response relationships, particularly the theoretical and practical similar ities and differences between exposure to chemical carcinogens compared to exposure to ionizing radiation.
