Ce roman fait suite au Mystère de la chambre jaune. Après avoir été longtemps son soupirant, Robert Darzac peut enfin épouser Mlle Stangerson, la victime du mystérieux attentat de la chambre jaune. Mais l'agresseur court toujours, et c'est maintenant Robert Darzac qu'il tente d'assassiner. On appelle à l'aide le jeune reporter Rouletabille qui avait résolu le mystère du roman précédent. Pour y parvenir il était allé enquêter en Amérique où il a passé son enfance. Il y a identifié l'anonyme dame en noir qui lui rendait parfois visite à l'orphelinat et dont le parfum obsède sa mémoire. Aurait-elle un lien avec ces affaires criminelles ?
