Cette thèse porte sur la mise en œuvre et le démarrage d'un réacteur à lit turbulé inverse avec des particules d'ExtendospheresÔ pour le traitement anaérobie d'un effluent agro-alimentaire. Une étude hydrodynamique a permis de caractériser les écoulements et le macro-mélange en présence de ce support de petite taille (147æm) et de faible densité (0,7). L'hypothèse d'un pseudo-fluide homogène liquide-solide dont la viscosité apparente dépend de la quantité de solide illustre bien le comportement des phases lors de l'injection du gaz pour la fluidisation. Une étude biologique a montré que le contact initial entre cellules et support entraîne une adaptation physiologique des microorganismes à la présence de solide après une inhibition temporaire de la méthanisation. Le rendement en méthane s'est avéré un paramètre de suivi intéressant de la formation et du détachement du biofilm. Un faible temps de séjour hydraulique lors du démarrage a permis de réduire la phase de latence lors de la colonisation du support. Un fonctionnement continu robuste du réacteur a été obtenu grâce à un contrôle de l'alimentation par le pH. Elle a aussi permis d'étudier l'influence de la fluidisation par le gaz sur le taux d'attrition du biofilm. La vitesse du gaz est apparue comme un paramètre de contrôle actif de la proportion de cellules, de la densité et de la résistance du biofilm.
