Cette thèse se décompose en deux parties. Elle est consacrée à la résolution numérique de divers problèmes non-linéaires à l'aide d'une méthode de continuation. La première partie traite d'un problème de diffusion-réaction dans une membrane enzymatique chargée. La résolution de ce problème de perturbation singulière est délicate et couteuse par les moyens traditionnels. L'étude asymptotique du problème permet de dégager un problème non perturbé dont la résolution est plus aisée et conduit sans difficulté à une approximation de la différence de potentiel dans la membrane. La résolution de ce problème en fonction de divers paramètres est facilitée par l'emploi d'une méthode de continuation. La seconde partie, quant à elle, est consacrée à la résolution de quelques problèmes d'optimisation dans des systèmes à état multiples. Nous utilisons une méthode de minimisation classique pour rechercher soit un point limite, soit un point de bifurcation de Hopf sur une branche d'états stationnaires d'un système différentiel. Le contrôle dans les systèmes distribués est ensuite abordé par une méthode de pénalisation ou à l'aide d'une méthode d'optimisation déduite d'une méthode de continuation. Enfin l'optimisation d'un point de doublement de période a été réalisée par cette dernière méthode.
