Traffic jam (tj) encodes a soma-specific Maf transcription factor required for morphogenesis of the gonads in Drosophila melanogaster. I examined the requirement of tj function throughout oogenesis by studying the expression of follicle cell type specific markers in tj mutant cell clones. I also examined the interaction between tj and JAK-Stat pathway genes using a variety of loss- and gain-of-function mutations. As well as regulating the expression of a number of adhesion molecules, TJ appears to be a general requirement in follicle cells for their proper differentiation throughout oogenesis. Depending on when cells lose tj, their subsequent differentiation is blocked or defective. This suggests that like other Mafs, TJ is a regulator of differentiation. Furthermore, my findings suggest that TJ interacts with the JAK-Stat pathway during oogenesis. I have also identified a possible interaction between TJ and the EGF receptor signaling pathway.

The gene traffic jam (tj) encodes a soma-specific, large Maf transcription factor, required for gonad morphogenesis in Drosophila. My gain-of-function analysis of tj in adult ovaries revealed both, defects in cell specification and cell behaviour. The ectopic expression of tj during oogenesis produced follicles with ectopic polar cells, missing inter-follicular stalks and stalk cells, absent or non-migrating border cells, non-migrating centripetal cells and defective stretch cells. These tj gain-of-function phenotypes mimic the phenotypes found in Jak-Stat signalling pathway mutants. My data suggest that TJ may normally be involved in cell specification, and may interact with the Jak-Stat signalling pathway. It was also found that two of the mouse homologues of tj, MafB and c-Maf, are expressed in somatic tissues of adult mouse gonads, suggesting that the role of tj in gonad development and gametogenesis may be evolutionarily conserved.

DURING MY PHD WORK, I INVESTIGATED THE ROLE OF THE SMALL MONOMERIC GTPASE RAB6 IN ORGANIZATION OF THE EGG-CHAMBER AND ASYMMETRIC LOCALIZATION OF THE BODY AXIS DETERMINANTS DURING OOGENESIS. I FOUND THAT ABSENCE OF RAB6 FUNCTION CAUSES EGG-CHAMBER DISORGANIZATION AND LOSS OF NURSE CELL PLASMA MEMBRANES. I ALSO OBSERVED THAT EXOCYTOSIS IS AFFECTED IN RAB6 NULL EGG-CHAMBERS, PROVIDING AN EXPLANATION FOR THE OBSERVED PHENOTYPES. RAB6 IS ALSO REQUIRED FOR POLARIZATION OF THE OOCYTE MICROTUBULE CYTOSKELETON AND LOCALIZATION OF OSKAR MRNA AT THE OOCYTE POSTERIOR POLE. FURTHERMORE, I SHOWED THAT, IN VIVO, RAB6 IS FOUND IN A COMPLEX WITH BICAUDAL-D, KNOWN FOR ITS ROLE IN MRNA LOCALIZATION IN DROSOPHILA. GENETIC ANALYSIS REVEALS THAT, IN THE OOCYTE, RAB6 AND BICAUDAL-D COOPERATE IN OSKAR MRNA LOCALIZATION. AS A CONCLUSION, I PROPOSE THAT DURING DROSOPHILA OOGENESIS, RAB6-DEPENDENT MEMBRANE TRAFFICKING IS DOUBLY REQUIRED, FIRST FOR GENERAL ORGANIZATION AND GROWTH OF THE EGG- CHAMBER, AND SECOND, MORE SPECIFICALLY, FOR POLARIZATION OF THE MICROTUBULE CYTOSKELETON AND LOCALIZATION OF OSKAR MRNA. Life Science studies in space were initially driven by the need to explore how man could survive spaceflight conditions; the effects of being launched un der high accelerations, exposed to weightlessness and radiation for different periods of time, and returned to Earth in safety. In order to substantiate the detailed knowledge of potentially adverse effects, many model experiments were launched using organisms which ranged from bacteria, plants, inverte brates, rodents and primates through to man. Although no immediate life threatening effects were found, these experiments can be considered today as the precursors to life science research in space. Many unexplained effects on these life forms were attributed to the condition of weightlessness. Most of them were poorly recorded, poorly published, or left simply with anecdotal information. Only with the advent of Skylab, and later Spacelab, did the idea emerge, and indeed the infrastructure permit, weightlessness to be considered as an ex tended tool for research into some fundamental mechanisms or processes as sociated with the effect of gravity on organisms at all levels. The initial hy pothesis to extrapolate from hypergravity through 1 x g to near 0 x g effects could no longer be retained, since many of the experiment results were seen to contradict the models or theories in the current textbooks of biology and physiology. The past decade has been dedicated primarily to exploratory research.
