The fourth annual American Institute for Cancer Research (AICR) conference on diet, nutrition and cancer was held at the L'Enfant Plaza Hotel in Washington, D. C. , September 2~3, 1993. In keeping with present concerns and in line with current trends, the theme was "Diet and Breast Cancer. " This proceedings volume is comprised of chapters from the platform presentations of the two day conference and abstracts from the poster session held at the end of the first day. Experimentally, there is sufficient evidence to support a relationship between dietary fat and the risk of breast cancer. A meta-analysis was provided by data from 114 experiments with over 10,000 animals, divided into groups fed ad libitum on diets with different levels or sources of fat, or different levels of energy restriction. This exercise suggested that linoleic acid was a major determinant of mammary tumor development but that other fatty acids also enhanced mammary tumor development in animals. However, as mentioned by several speakers, results from epidemiological studies often are conflicting, thus leading to confusion among both health professionals and the public. Surveys of specific populations which have migrated from countries with low breast cancer rates to those with higher rates are often some of the most compelling studies with respect to a high fat diet-breast cancer association. Nonetheless, various cohort and prospective studies, some quite large, did not appear to show a relationship between consumption of fat (any type) and breast cancer.

The American Institute for Cancer Research (AICR) sponsored its third annual confer ence on nutrition and cancer. The theme was "Diet and Cancer: Markers, Prevention, and Treatment. " The conference was held October 29-30, 1992 at the Ritz Carlton Hotel in McLean, Virginia. This proceedings contains chapters from the platform presentations and abstracts from the poster presentations. Several chapters address each of four session topics: Retinoids as Differentiation Agents in Cancer Therapy, Biological Markers of Cancer Risk, Chemoprevention of Cancer by Non-Nutrients in Foods, and Nutritional Problems and Support in the Treatment of Cancer. The first three chapters discuss in detail different mechanisms by which retinoids influence differentiation and provide evidence to support their use in cancer therapy. In vivo and in vitro studies show the effects of retinoic acid (RA) on tumorigenicity and cellu lar/molecular events. A synopsis of data showing the involvement of the retinoblastoma (RB) gene in HL-60 cell differentiation induced by RA and 1,2S-dihydroxy vitamin D3 (VD) is presented. In SCID (severe combined immunodeficiency) mice injected with HL-60 3 human leukemia cells and gavaged daily with RA, the number of tumor sites and number of mice with tumors are reduced. All trans-RA induces myeloid differentiation in HL-60 cells. Similarly, VD3 induces HL-60 monocytic differentiation. In both cases an early down regulation of retinoblastoma (RB) gene expression precedes the differentiation.

The sixth annual research conference of the American Institute for Cancer Research was held August 31 and September 1, 1995, at the Loews L'Enfant Plaza Hotel in Washing ton, DC. In view of the promising leads in the diet/nutrition and cancer research field, the conference was devoted to "Dietary Phytochemicals in Cancer Prevention and Treatment. " The number of sessions was increased over that in previous conferences in order to accommodate the topics of interest. The conference overview, entitled "Plants and Cancer: Food, Fiber, and Phytochemicals," provided a framework for the following sessions. In addition, the attendees were reminded that for several decades epidemiologists have noted a lower risk of lung, esophageal, stomach, and colon cancer in populations consuming diets high in fruits and vegetables. However, isolation and ingestion of individual protective factors are not the preferred action since the complexity of the food and the matrix in which nutritional factors are embedded are important. The individual sessions then provided more insight as to why eating fruits and vegetables is associated with a lower risk of cancer. The first of these sessions was on "Isothiocyanates" that induce both the Phase I and Phase II enzymes that increase detoxification and conjugation reactions, thus causing more rapid removal of any xenobiotic or carcinogen. Thus, less carcinogen is available for interaction with DNA or other critical cellular macromolecules.
